  • Resumo Summary

    Nosso grupo de pesquisa está focado na patogenia da cardiopatia chagásica crônica (CCC). Esta é uma doença endêmica que afeta milhões de pessoas em toda a América Latina. Estudos anteriores mostraram segregação de variantes genéticas raras em genes mitocondriais e inflamatórios com os casos de CCC.

    Nosso grupo combina métodos de biologia celular e molecular, imunologia, genética e bioenergética. Estamos interessados em estudar o sinergismo entre o dano mitocondrial causado por tais variantes genéticas e a alta produção de interferon-gama (IFN-γ) observada nos pacientes com CCC.

    Os bolsistas selecionados no âmbito desta oportunidade irão explorar a fisiopatologia de mitocôndrias e citocinas na CCC, realizando estudos funcionais com 1) cardiomiócitos derivados de células-tronco pluripotentes induzidas obtidas de pacientes, portadores ou não das variantes genéticas acima citadas; e 2) linhagem permanente de cardiomiócitos editadas por CRISPR/Cas9 carregando ou não as variantes genéticas de interesse, sob o efeito de IFN-γ e TNF-α. Esses estudos incluem a avaliação da função mitocondrial e alterações transcriptômicas em cardiomiócitos tratados ou não com citocinas.

    A vaga está aberta a brasileiros e estrangeiros. Cada selecionado receberá Bolsa de Pós-Doutorado da FAPESP, válida por dois anos, no valor de R$ 8.479,20 mensais, que inclui ainda Reserva Técnica equivalente a 10% do valor anual da bolsa para atender a despesas imprevistas e diretamente relacionadas à atividade de pesquisa.

    The pathogenesis of Chagas disease cardiomyopathy (CCC), an endemic disease affecting millions of patients in Latin America, is the focus of Dr. Cunha-Neto’s group at the Heart Institute (InCor) of Hospital das Clínicas (HC), the hospital complex run by the Medical School of the University of São Paulo (FM-USP) in Brazil. Previous studies have disclosed rare heterozygous pathogenic variants in mitochondrial and inflammatory genes segregating in CCC cases.

    We are interested in studying the synergy in mitochondrial damage caused by such genetic variants and the high interferon-gamma (IFN-γ) production observed in CCC. Our group combines molecular/cell biology, immunology, genetics and bioenergetics methods.

    This opportunity will select candidates with aim of exploring the pathophysiology of mitochondria and cytokines in CCC by performing functional studies with 1) induced pluripotential stem cell (iPSC)-derived cardiomyocytes from the patients carrying or not the genetic variants of interest, as well as 2) CRISPR-Cas9 gene-edited cardiomyocyte cell lines carrying or not the genetic variants of interest, under the effect of IFN-γ and TNF-α. These studies include mitochondrial function and transcriptomic changes in cytokine-treated cardiomyocytes.

    This opportunity is open to candidates of any nationality. Each selected candidate will receive a Post-Doctoral fellowship from the São Paulo Research Foundation (FAPESP) in the amount of R$ 8,479.20 monthly for 2-4 years, and a research contingency fund equivalent to 10% of the annual value of the fellowship which should be spent on items directly related to the research activity.
