  • Resumo Summary

    Nossos resultados revelaram que a exposição a cobra induziu em camundongos uma expressão exuberante da proteína Fos na região do núcleo Edinger-Westphal (EW), particularmente localizada na parte de projeção central do EW (EWcp). É importante ressaltar que o EWcp é um dos principais sítios que expressam a urocortina 1 (Ucn1) e a transcrição regulamentada pela cocaína e anfetamina (CART). Verificou-se que a EWcp está envolvida no controle da resposta ao estresse e na ingestão de alimentos, além de regular o consumo de álcool e facilitar a nocicepção.

    No presente caso, seria importante entender como o EWcp influencia as respostas comportamentais e de estresse no contexto da exposição a cobra. Para esse fim, realizaremos um estudo de rastreamento de vias neurais para examinar as projeções eferentes do EWcp. Para abordar os neurônios EWcp, usaremos camundongos transgênicos que expressam a recombinase de CRE nos neurônios CART e aplicaremos a sequência flox expressa no vírus adeno-associado para marcar com EYFP as fibras de projeção de neurônios EWcp específicos. Em seguida, realizaremos estudos funcionais de EWcp no contexto da exposição a serpentes.

    O candidato deve ter experiência em rastreamento de vias neurais e análise comportamental e enviar um breve currículo e uma carta de interesse explicando a motivação e a experiência anterior para realizar o projeto.

    A vaga está aberta a brasileiros e estrangeiros. O selecionado receberá Bolsa de Pós-Doutorado da FAPESP no valor de R$ 7.373,10 mensais e Reserva Técnica equivalente a 15% do valor anual da bolsa para atender a despesas imprevistas e diretamente relacionadas à atividade de pesquisa.

     

    Results of research carried out within the present project, based in the Biomedical Sciences Institute of the University of São Paulo (ICB-USP), revealed that exposure to snake induced in mice an exuberant expression of Fos protein in the region of the Edinger-Westphal (EW) nucleus, particularly located in the central projection part of the EW (EWcp). Importantly, EWcp is one of the main sites that express urocortin 1 (Ucn1) and the transcript regulated by cocaine and amphetamine (CART). It was found that EWcp is involved in controlling the stress response and food intake, in addition to regulating alcohol consumption and facilitating nociception.

    In the present case, it would be important to understand how EWcp influences behavioral and stress responses in the context of snake exposure. To that end, we will carry out a neural pathway tracking study to examine the efferent projections of the EWcp. To address EWcp neurons, we will use transgenic mice that express CRE recombinase in CART neurons and apply the flox sequence expressed in adeno-associated virus to mark with EYFP the projection fibers of specific EWcp neurons. Next, we will carry out functional studies of EWcp in the context of snake exposure.

    For this project, the candidate must have good experience in neural track tracing and behavioral analysis. Applicants must send a short CV and a letter of interest explaining the motivation and previous experience to carry out the project.

    This opportunity is open to candidates of any nationalities. The selected candidate will receive a Post-Doctoral fellowship from the São Paulo Research Foundation-FAPESP in the amount of R$ 7,373.10 monthly and a research contingency fund, equivalent to 15% of the annual value of the fellowship which should be spent in items directly related to the research activity.
