    Oportunidade aberta para bolsa FAPESP de Pós-Doutorado no âmbito do projeto "Desenvolvimento e padronização de uma nova metodologia para avaliar o efeito de peptídeos intracelulares em interações proteína-proteína". A pesquisa será desenvolvida na Universidade de São Paulo, Instituto de Ciências Biomédicas, Departamento de Farmacologia, situado na Av. Lineu Prestes 1524, Sala 317, São Paulo, SP, 05508-000, Brasil.

    O projeto está vinculado ao Projeto Temático "Farmacologia de oligopeptidases e peptídeos intracelulares/Pharmacology of oligopeptidases and intracellular peptides" (Processo #2016/04000-3).

    Nosso objetivo nesse projeto será desenvolver e validar um protocolo experimental que permita a quantificação de interações entre proteínas in vivo (em células e/ou animais), que possa ser aplicado para determinarmos o efeito de peptídeos intracelulares específicos, ou de alterações globais na composição intracelular de peptídeos.

    Buscamos indivíduos qualificados e capazes de combinar habilidades em farmacologia molecular e bioquímica com a capacidade de trabalhar em equipe e liderar projetos científicos no campo.

    Os candidatos devem ter Doutorado ou PhD em Farmacologia, Bioquímica, Biologia Celular ou Biologia Molecular, com foco em estudos moleculares e celulares que visem a descoberta de novos fármacos e novas vias de sinalização celular.

    Além disso, é necessário experiência em farmacologia, análise de sistemas celulares de sinalização como dosagem de segundos mensageiros e avaliações de atividade enzimática. Conhecimentos de cromatografia liquida e espectrometria de massas são desejados.

    O candidato contribuirá com análise e integração dos resultados gerados para publicação em revistas de alto impacto. Nível avançado de inglês para escrita e conversação é obrigatório. O candidato ideal será aquele apto a contribuir na supervisão de estudantes de pós-graduação.

    O candidato dever ter registro de publicações em revistas especializadas de qualidade, indexadas no Pubmed, e de preferência de impacto igual ou superior a dois (2).

    Deve ter disponibilidade imediata para residir em São Paulo. 

    Caso seja selecionado, o candidato terá uma bolsa de Pós-Doutorado da FAPESP (www.fapesp.br/bolsas/pd) no valor mensal de R$ R$ 7.174,80 por 24 meses (renovável por até mais 12 meses).

    Resumo do Projeto

    As proteínas são fundamentais para a dinâmica fisiológica da vida, formando redes interativas que respondem às circunstâncias espaciais e temporais (por exemplo, condições ambientais, estresse ou situações patológicas ou à ação de drogas). As proteínas não agem sozinhas, mas interagem entre si, participando de uma série de modulações fisiológicas. Estas interações proteína(s)-proteína(s) podem ser dinâmicas ou estáveis, e ocorrem através de mecanismos estruturais físico-químicos, controlados pelo microambiente estrutural e celular. Devido à dinâmica e complexidade destas reações, o ideal ao serem analisadas é que os métodos analíticos utilizados perturbem minimamente o contexto natural celular.

    Dentre a vasta variedade de proteínas e peptidases sugeridas regulando interações proteína-proteína encontramos a thimet oligopeptidase (EC 3.4.24.15; EP24.15). Essa enzima se destaca pela presença em ambos os compartimentos intracelular e extracelular, onde metaboliza diversos peptídeos (degrada e produz) produzidos pelo proteassomo ou pela via secretória (i.e. neuropeptídeos). A EP24.15 participa da rede de interações celulares interagindo, por exemplo, com a 14-3-3 epsilon de forma dependente da fosforilação na serina 644 pela proteína quinase A (Carreño ET AL., 2005). Por outro lado, a EP24.15 também foi sugerida participar da rede de interações proteicas celulares metabolizando peptídeos intracelulares (Cunha ET AL., JBC 2008; Russo ET AL., FEBS Letters, 2012).

    Diante disso, nosso desafio nesse projeto é desenvolver e validar um protocolo experimental que permita a quantificação de interações entre proteínas in vivo (em células e/ou animais), que possa ser aplicado para determinarmos o efeito de peptídeos intracelulares específicos, ou de alterações globais na composição intracelular de peptídeos.

    Opportunity opened for a post-doctoral fellowship granted by the Sao Paulo Research Foundation -- FAPESP to work in the project "Development and standardization of a new methodology to evaluate the effect of intracellular peptides on protein-protein interactions". The project will be conducted at the University of São Paulo, Institute of Biomedical Sciences, Department of Pharmacology, located at Av. Lineu Prestes 1524, Room 317, São Paulo, SP, 05508-000, Brazil.

    The project is linked to the Thematic Project (Grant number 2016/04000-3) "Pharmacology of oligopeptidases and intracellular peptides". 

    Our objective in this project will be to develop and validate an experimental protocol that allows the quantification of interactions between proteins in vivo (in cells and / or animals), that can be applied to determine the effect of specific intracellular peptides, or of global changes in intracellular composition of peptides. 

    We are looking for qualified individuals who are able to combine skills in molecular pharmacology and biochemistry with the ability to work as a team and lead scientific projects in the field.

    Applicants must have a PhD or PhD in Pharmacology, Biochemistry, Cell Biology or Molecular Biology, focusing on molecular and cellular studies aimed at discovering new drugs and new cell signaling pathways.

    In addition, experience in pharmacology, analysis of cellular signaling systems as a second messenger dosage and enzyme activity assays are required. Knowledge of liquid chromatography and mass spectrometry are desired.

    The candidate will contribute with analysis and integration of the results generated for publication in high impact journals. Advanced level of English for writing and speaking is required. The ideal candidate will be the one able to contribute to the supervision of postgraduate students.

    The candidate must have a record of publications in specialized quality journals, indexed in Pubmed, and preferably with an impact of two or more (2).

    Must have immediate availability to reside in São Paulo. 

    If selected, the candidate will have a post-doctoral fellowship from FAPESP (www.fapesp.br/en/5427) in the monthly amount of R$ 7,174.80 per month, during 24 months (renewable for up to 12 months). The fellowship also includes a research contingency fund, equivalent to 15% of the annual value of the fellowship which should be spent in items directly related to the research activity.

    Project Summary

    Proteins are fundamental to the physiological dynamics of life, forming interactive networks that respond to spatial and temporal circumstances (eg, environmental conditions, stress or pathological situations or drug action). Proteins do not act alone, but interact with each other by participating in a number of physiological modulations. These interactions protein (s) -protein (s) can be dynamic or stable, and occur through physical-chemical structural mechanisms, controlled by the structural and cellular microenvironment. Due to the dynamics and complexity of these reactions, the ideal when analyzed is that the analytical methods used will disturb the natural cellular context.

    Among the vast variety of proteins and peptidases suggested to regulate protein-protein interactions are thimet oligopeptidase (EC 3.4.24.15; EP24.15). This enzyme is notable for its presence in both the intracellular and extracellular compartments, where it metabolizes various peptides (degraded and produced) produced by the proteasome or the secretory pathway (i.e., neuropeptides). EP24.15 participates in the network of cellular interactions interacting, for example, with 14-3-3 epsilon in a manner dependent on phosphorylation at serine 644 by protein kinase A (Carreño ET AL., 2005). On the other hand, EP24.15 has also been suggested to participate in the network of cellular protein interactions by metabolizing intracellular peptides (Cunha ET AL., JBC 2008; Russian ET AL., FEBS Letters, 2012).

    Therefore, our challenge in this project is to develop and validate an experimental protocol that allows the quantification of interactions between proteins in vivo (in cells and / or animals), that can be applied to determine the effect of specific intracellular peptides, or of global changes in the intracellular composition of peptides.
