    O Laboratório de Fisiologia do Departamento de Fisiologia e Patologia da Faculdade de Odontologia de Araraquara – Unesp tem uma vaga em aberto para Bolsa de Pós-Doutorado da FAPESP na área de Fisiologia Cardiovascular e Respiratório. 

    A bolsa está vinculada ao Projeto Temático coordenado pelo Prof. Dr. Eduardo Colombari intitulado “Fisiopatologia experimental: Mecanismos centrais de controle cardiovascular erespiratório envolvidos em modelos experimentais de hipertensão e obesidade”.

    Resumo:

    O Projeto Temático visa à compreensão dos mecanismos neurais que processam informações autonômicas associadas aos ajustes cardiovasculares e respiratórios subjacentes à produção de hipertensão arterial em diferentes modelos experimentais. O foco do estudo será sobre um modelo de circuito que envolve o triângulo formado por regiões romboencefálicas, núcleo do trato solitário (NTS), região rostro-ventrolateral do bulbo (RVL) e suas conexões com regiões do prosencéfalo como lâmina terminal e núcleo paraventricular do hipotálamo.

    Os objetivos do projeto são:

    1) Estudar o envolvimento do sistema reninaangiotensina e o sistema melanocortina centrais nas alterações cardiovasculares e ventilatórias induzidas pela obesidade;

    2) Estudar se os mecanismos neurais de controle basal e reflexo das atividades simpática e respiratória são susceptíveis à plasticidade em resposta a alterações no aporte de nutrientes durante a fase de desenvolvimento/maturação;

    3) Estudar os efeitos do treinamento resistido como medida preventiva para a correção das alterações cardiovasculares em ratos submetidos à dieta hiperlipídica;

    4) Verificar a participação do estresse oxidativo em diferentes áreas encefálicas para o desenvolvimento/manutenção em um modelo animal de hipertensão renovascular (2-rins, 1-clipe, 2R1C) e essencial (ratos espontaneamente hipertensos, SHR);

    5) Estudar a inter-relação entre o sistema reninaangiotensina central e as citocinas inflamatórias no sistema nervoso central como um possível mecanismo facilitatório para o desenvolvimento de diferentes modelos de hipertensão (2R1C, SHR e induzida pela obesidade).

    Para atingir esses objetivos o projeto combinará diversas técnicas tais como: injeções intraencefálicas de fármacos; transfecção viral; imunohistoquímica; análise da expressão gênica quantitativa, eletrofisiologia; registros cardiovasculares e ventilatórios.

    O candidato a ser selecionado deverá ter sólida formação em eletrofisiologia, com experiência demonstrada em registro de nervos em preparações isoladas e de neurônios (patch-clamp) em fatias cerebrais e/ou em preparações in situ. Experiência consolidada na análise e processamento de sinais biológicos em softwares de uso rotineiro em laboratórios de eletrofisiologia é condição indispensável para os candidatos a essa bolsa de pósdoutoramento.

    A seleção será por meio de análise do currículo, seminário e entrevista. A bolsa tem valor de R$ 6.819,30/mês e duração de até 24 meses, dependente de análise de mérito pela FAPESP.

    É desejável fluência em inglês.     The Department of Physiology and Pathology of the Dentistry School of Araraquara - UNESP has an opened pos-doc position with scholarship from FAPESP to work with Cardiovascular and Respiratory Central Nervous System Control.

    The position is tied with the Thematic Project “Experimental pathophysiology: Role of central mechanisms of the cardiovascular and respiratory changes induced by experimental hypertension and obesity”, coordinated by Professor Eduardo Colombari.

    Abstract:

    Obesity and hypertension are the leading cause of death in the world. Obesity and overweight contribute to elevated blood pressure in many patients with essential hypertension. Experimental and clinical studies indicate that visceral obesity induces hypertension by increasing renal sodium reabsorption through multiple mechanisms including activation of the sympathetic nervous system (SNS). Obesity-related hypertension is associated with altered responsiveness of SNS and the renin-angiotensin-system (RAS), possibly leading to blunted sympatho-inhibitory and vasodilator effects in the renal and splanchnic vasodilator beds.

    The present Thematic Project aims to understand the neural mechanisms that process such neuroautonomic information associates to the cardiovascular and respiratory adjustments during the development of hypertension in different experimental models.

    The main focus of this study is a model of circuit that involves the triangle formed for rhomboencephalic regions, nucleus of the solitary tract (NTS), rostroventrolateral region of the medulla (RVLM) and its connections with prosencephalic regions like lamina terminalis and paraventricular nucleus of the hypothalamus.

    From the candidate to work on this project it would be expected that the person know how to combine different techniques such as: central injections of drugs; viral transfection; imunohystochemistry; analysis of the quantitative gene expression, electrophysiology; cardiovascular and ventilatory measurements. 

    To be selected the candidate must have solid education on electrophysiology, with experience demonstrated in both nerve and neurons recording in cerebral slices and preparations in situ.

    Experience consolidated in the analysis and processing of biological signals in software of routine use in electrophysiology laboratories is indispensable condition for the candidates to this position.

    The post-doc scholarship is about R$ 6.819,30/month free of tax and the duration is up to 24 months.

    Must be fluent in English.
