Summary
Mecanismos fotoinduzidos de morte celular programada e os diferentes perfis de expressão gênica.
A terapia fotodinâmica (PDT/Photodynamic therapy) é um tratamento contra diversas enfermidades humanas que tem como princípio básico o emprego de uma molécula fotossensível que, uma vez internalizada pela célula e excitada por radiação de luz visível (600-800nm), gera diversas moléculas reativas e induz a morte do tecido alvo.
O objetivo principal do projeto temático em vigência é identificar mecanismos de aumento de especificidade na indução de morte celular fotoinduzida.
Em estudos prévios, caracterizamos maneiras de induzir, com fotossensibilizadores semelhantes, mecanismos distintos de morte celular programada. Esta indução mostrou-se dependente, principalmente, da localização celular do fotossensibilizador.
Embora o foco do laboratório tenha sempre sido em células eucarióticas, pretendemos, nos próximos anos, também aplicar estes conceitos em organismos procariotos.
Neste projeto pretendemos comparar, qualitativamente e quantitativamente, a expressão gênica induzida pela fotossensibilização em função da localização celular do fotossensibilizador e do tipo de morte celular programada induzida. Além disso, expandiremos esses estudos para organismos procariotos.
Características do candidato: conhecimento/experiência em bioquímica celular, microbiologia, biologia celular e molecular, processos patológicos e cultura de células.

Photodynamic therapy (PDT/Photodynamic therapy) is a treatment against several human diseases whose basic principle is the use of a photosensitive molecule, which is internalized by the cell and excited with visible light (600-800nm), generating several reactive species and inducing cell death in the target tissues.
The main objective of the thematic research grant is to identify mechanisms to induce programed cell death, using light.
In previous studies, we characterized ways to induce, with similar photosensitizers, distinct mechanisms of programmed cell death. This induction was shown to be dependent mainly on the cellular localization of the photosensitizer.
Although the lab focus has always been in eukaryotic cells, we intend, in the coming years, to also apply these concepts in prokaryotes organisms.
In this project we will compare qualitatively and quantitatively the gene expression induced by the photosensitivity reactions depending on the cellular localization of the photosensitizer and on the type of programmed cell death. Furthermore, we also aim to expand these studies to prokaryotic organisms.
Characteristics of the candidate: knowledge/experience in cellular biochemistry, microbiology, molecular and cell biology, pathological processes and cell culture.

This opportunity is open to candidates of any nationalities.
