    Uma vaga de pós-doutorado, com duração de até 48 meses, está disponível no Departamento de Clínica Médica da Faculdade de Medicina de Ribeirão Preto (FMRP) da Universidade de São Paulo (USP), em Ribeirão Preto (SP). O pesquisador irá desenvolver o projeto intitulado: “Genotipagem de T. cruzi em amostras de pacientes com Cardiopatia Crônica da Doença de Chagas. Desenvolvimento de capacidade tecnológica na FMRP-USP. Para tal, o pesquisador selecionado a partir deste edital deverá desempenhar suas atividades científicas no âmbito do Laboratório de Imunoparasitologia liderado pelo Professor João Santana da Silva.

    Os candidatos devem ser excepcionalmente motivados, inovadores, ser capazes de trabalhar de forma independente em ambiente de equipe. O candidato ideal deve ter um grau de doutoramento e formação nas áreas de Biologia Celular e Molecular, Bioquímica ou Doenças Infecciosas. É necessária experiência prévia em técnicas como Biologia Molecular, Bioquímica e Imunoparasitologia. Ter experiência em variabilidade genética do Trypanosoma cruzi, genotipagem e suas DTU's.

    Resumo do subprojeto, inserido no contexto de um projeto temático aprovado pela FAPESP sob a coordenação do Prof. José Antônio Marin-Neto, intitulado: "Investigações sobre etiopatogenia, fisiopatologia e terapêutica em humanos e em modelo experimental com a cardiomiopatia da Doença de Chagas".

    Resumo do projeto:

    O Trypanosoma cruzi possui estrutura que permite subdivisão em seis grupos genéticos distintos, denominados de TcI a TcVI, com diferenças em termos de distribuição geográfica, propriedades biológicas e susceptibilidade a medicamentos. A correlação entre a variabilidade genética parasitária e as formas clínicas da Doença de Chagas (DCh), permanece incompletamente estabelecida. É conhecido o predomínio das DTU’s (Grupos genéticos) TcII e TcVI em diversas regiões brasileiras, associados tanto ao ciclo doméstico como ao silvestre da DCh.

    Tenciona-se com este projeto verificar os genótipos dos parasitos presentes em 100 pacientes com DCh arrolados em Ribeirão Preto, e avaliar a existência de potenciais alvos no genoma dos parasitos, a fim de correlacionar os resultados com a evolução clínica dos pacientes dos quais são provenientes os parasitos.

    Além da genotipagem, será realizada triagem genômica dos parasitos, isolados dos pacientes por hemocultura, para identificação de genes potenciais alvos, que sirvam como marcadores de prognóstico para a infecção da DCh. Esse objetivo será alcançado simultaneamente à consecução da meta primordial, qual seja, de dotar o laboratório chefiado pelo Professor João Santana da Silva, pesquisador supervisor dos trabalhos a que se refere este sub-projeto, do Departamento de Bioquímica e Imunologia, da FMRP-USP, de capacidade de genotipagem de T. cruzi para aproveitamento ulterior em outros projetos de aplicação clínica. 

    A 48-month postdoctoral position is available at the Department of Internal Medicine of the Ribeirão Preto Medical School of the University of São Paulo (USP), Ribeirão Preto, São Paulo State, Brazil. The researcher will develop the project titled: "Genotyping of Trypanosoma cruzi in samples from patients with Chronic Chagas' Heart Disease’’.

    Development of scientific capability in the Medical School of Ribeirão Preto, USP. To do so, the Laboratory of Imunoparasitology, led by Professor João Santana Silva, is seeking for a highly motivated Post-Doctoral individual. The Laboratory is located in the Department Biochemistry and Immunology, Ribeirão Preto Medical School from the University of Sao Paulo (FMRP-USP), Ribeirão Preto, Brazil. Successfully applicants can be funded for up to 4 years by the São Paulo Research Foundation-FAPESP.

    Applicants must be exceptionally motivated, innovative, that should be able to work independently and in a collaborative team setting. The ideal candidate must have a PhD degree and background in the fields of Cell Biology, Biochemistry or Infectious Diseases. Prior experience in techniques such as Molecular Biology, Biochemistry and Immunoparasitology is necessary. Have experience in genetic variability of Trypanosoma cruzi, genotyping and DTU's.

    Summary of the subproject, that is part of the thematic project supported by FAPESP, under coordination of Prof. José Antônio Marin-Neto, entitled: Research on etiopathogeny, physiopathology and therapeutics in humans and experimental models of Chagas disease cardiomyopathy. 

    Abstract of Project

    Trypanosoma cruzi has a structure that allows subdivision into six distinct genetic groups, called TcI to TcVI, with differences in terms of geographical distribution, biological properties and susceptibility to drugs. The correlation between parasitic genetic variability and clinical forms of Chagas disease (ChD) remains incompletely established. The predominance of DTUs (Genetic Groups) TcII and TcVI in several Brazilian regions, associated to both the domestic and wild cycle of ChD is well known.

    This project intends to verify the genotypes of the parasites present in 100 patients with ChD enrolled in Ribeirão Preto and to evaluate the existence of potential targets in the parasite genome in order to correlate the results with the clinical evolution of the patients from whom the parasites were isolated. In addition to genotyping, genomic screening of the parasites, isolated from the patients by blood culture, will be carried out to identify potential target genes, which serve as prognostic markers for human ChD.

    This objective will be achieved simultaneously to achieve the primary goal, that is, to equip the laboratory headed by Professor João Santana da Silva, researcher supervisor of the works referred to in this subproject, Department of Biochemistry and Immunology, FMRP-USP, of genotyping capability of T. cruzi for further use in other clinical application projects.

    This opportunity is open to candidates of any nationalities. The selected candidate will receive a FAPESP’s Post-Doctoral fellowship in the amount of BRL 7,174.80 monthly and a research contingency fund, equivalent to 15% of the annual value of the fellowship which should be spent in items directly related to the research activity.
